A lot of house music similar to this starts off with people sampling their favourite old funk/soul/jazz/disco records. There are no real shortcuts and I doubt if people are going to donate you their best ideas. You'll need a good record collection and a good ear.

Which of Live's instruments have you got loaded into the channel you are using?

Have you followed the tutorials that show you how to set up your sound card and your MIDI keyboard?
What's the urgency, do you need to have that hit record finished by tomorrow?

It is probably ground loop hum. Does the hiss stop when you unplug the laptop?

iTunes usually performs some 'audio enhancing' processing on the tunes it plays, such as bass maximising and harmonic exciting. It may make a finished mix sound better for some, but it's not something you'd want as a default when you're constructing a mix.
