If the effect has a Dry/Wet control, then just automate that envelope. Set it to 0% when you don't want the effect applying, and then jump it up to whatever you want during the sections you want it on.

Not sure about your first question. As for the second, have you tried automating the volume parameter for that one drum pad? First, expand the drum rack so you can see the controls for the particular sample/pad. Then show the envelopes, and use the automation to get that pad's volume to change as yo...
