Assuming you've got a loop mode seton the Sample tab, also make sure you have the sustain on the amp envelope up in the Filter/Global tab.

If you edit your audio with Sampler or Simpler you can enable Snap mode or use short fades to avoid pops.

Are your new Sends set to Pre-Fader? They should be if you want the audio from them to still hit the return tracks even with the source tracks' volumes down.

The short answer is probably "yes". However, you're going to need to be more specific about what you're trying to do. For starters, and LFO and a filter are two different things. What, exactly, are you trying to accomplish?
