audio pauses before instruments play

Whe I play a set the audio drops out and the set kind of pauses (just for a split second) when it gets to a midi part, that's playing an instrument like operator for example. This only happens at the first midi part and the first time through the set then when I play the set again it plays fine with no pauses.
I have tried playing with the sound card buffer and plug-in buffer settings with no success. So I have left them at 128 and 'As Audio Buffer'.
Anyone else experienced this? any solutions?
My system:
Windows XP Pro SP1
AMD Athlon 3700+ Novatech Laptop
1024 MB RAM
80 GB 5400rpm HD
M-Audio FW410 (5.10.0.5036)
Live 5.0.3
