It seems that Live needs to be re-booted in order for buffer size changes to take affect. Is there a way around this? Is there a way to re-launch the driver, for example? Or any way to change the buffer size without quiting the program?

That's your sound card's problem, not Live. My Edirol UA 101's buffers cannot be changed while Live is running, but my Echo Gina can, as well as the built in sound cards on my desktop (Creative shitblaster) and laptop.
