Parameter changes have no effect

I first thought this was a bug with sidechaining, but I've since realised it isn't. So I repost/rephrase:

Sometimes when you tweak parameters, they don't affect the sound even though the UI reflects the parameter changes.

I've noticed this three times, with ComboSister 1.40 (VSTi), Amplitube 2.1 (VST f/x) and L7's compressor.

ComboSister didn't seem to apply any changes (though the UI updated to reflect the changes I made).

The sound of Amplitube and Live's compressor both changed in response to a few of the parameters I tweaked but not to most of them. The UI still looked like all my changes were recieved.

In all cases, if I saved, closed and re-opened the Live set in question (three different sets), tweaking parameters had the expected effect.

This was all with Live 7.01.

In two cases, my Remote Zero SL was disconnected and I used my mouse to tweak parameters, in one case (the ComboSister incident), the SL was connected but it didn't matter if I used it or my computer mouse.
